Abstract
Background: The autoimmune illness recognized as (T1DM) causes weight loss, polyuria, polydipsia, and hyperglycemia along with the progressive death of β-cells. The disease primarily affects children and adolescents. The HLA genes might production a significant part in predisposing to type 1 diabetes. Interleukin-35 as a fellow of the IL-12 family that affects of immune modulation in a number of diseases, for instance (T1D), inflammatory bowel illness, Graves' illness, rheumatoid arthritis. Aim of Study: This study aimed to investigate the frequency of HLA-DR4/DR4 type in type 1 diabetes (T1D) and evaluate of interleukin-35 Level in patients compared with healthy group. Methods: Case-control training had been directed on (100) contributors, who attend to Endocrinology and Diabetes Center - Al-Hussein Teaching Hospital in Samawa city/Iraq throughout the dated as of 9/November 2023 to the end of October 2024.The participants were divided in to two group with aged (2-14) years old: First, patients group consist of (50) participants. Second, control group consisted of (50) participants without any history of chronic and autoimmune diseases, (3) milliliter blood sample were collected from each participant, then using (mPCR) multiplex polymerase chain reaction for detection of HLA-DR4/DR4 type and use ELISA technique for detect of IL-35 level. Statistical Software SPSS 26.0 was used. Results: HLA-DR4/DR4 has been identified in 24.0% (12 out of 50) in pediatric sick with T1D , while 8% (4 out of 50) in healthy group. The mean level of interleukin-35 were (8.76 ± 1.91 ng/ml) in pediatric sick with T1D , while (19.06 ± 2.90 ng/ml) in healthy group. Conclusions: The data indicate that a positively related HLA DR4/DR4 type with TID patients and decrease rank of IL-35 in T1D sick paralleled through controlling group.
